How do I read an insulin syringe for peptide dosing?

On a U-100 syringe, 100 units = 1mL, so 1 unit = 0.01mL. Draw back the plunger until the rubber stopper's flat edge aligns with your target unit number. What is concentration and why does it matter?

How to Reconstitute Peptides: Step-by-Step Guide

Supplies needed

Lyophilised peptide vial

Refrigerator

Gather and inspect

Check both vials for cracks, cloudiness in the BAC water, or discolouration. The peptide powder should be white or off-white. Wash hands thoroughly and work on a clean surface.

Swab the tops

Wipe the rubber septum of both the peptide vial and BAC water vial with a fresh alcohol swab. Allow to air-dry for 10–15 seconds before piercing — do not blow or fan dry.

Draw bacteriostatic water

Using a fresh syringe, draw the calculated volume of BAC water from the BAC water vial. For most peptides, 1–2mL is standard. Use the calculator above to determine the exact volume.

Inject down the vial wall at 45°

Tilt the peptide vial at 45 degrees. Slowly depress the plunger so the water runs down the inside wall of the vial, not directly onto the powder cake. This prevents denaturation from mechanical force.

Swirl — do not shake

Gently roll the vial between your palms in a slow circular motion until the powder fully dissolves. The solution should become clear and colourless. Never shake — agitation denatures peptide bonds.

Label and refrigerate

Write the peptide name, date reconstituted, concentration (mcg/mL), and vial size on the label using a permanent marker. Store immediately at 2–8°C, away from the refrigerator door. Discard after 28–30 days.

NAD+ requires BAC saline

NAD+ (nicotinamide adenine dinucleotide) is best reconstituted with bacteriostatic saline (0.9% NaCl with 0.9% benzyl alcohol) rather than standard BAC water. This improves stability and is isotonic, which reduces injection discomfort.

Nasal peptides need sterile saline

Peptides administered intranasally — including Semax, Selank, VIP, and Oxytocin — should be reconstituted in sterile saline (0.9% NaCl, no preservative) or sterile water. Benzyl alcohol is a mucosal irritant at nasal concentrations and should be avoided for intranasal use.

Always label the vial with the reconstitution date and discard after 28 days regardless of how much remains.
Store reconstituted vials upright in the main body of the refrigerator, not in the door (temperature fluctuates there).
Protect from light — a small piece of aluminium foil around the vial prevents photodegradation.
Never freeze a reconstituted solution; ice crystal formation during freezing mechanically damages peptide structure.
Keep lyophilised vials away from humidity — use desiccant packs in storage containers.
If the solution appears cloudy, discoloured, or has visible particulates after reconstitution, discard it.

10 Common Peptide Reconstitution Mistakes to Avoid

Confusing mg and mcg. One milligram equals 1,000 micrograms. A 5mg vial contains 5,000mcg. Always convert to the same unit before calculating. The calculator handles this automatically.
Confusing syringe units with International Units. On an insulin syringe, “units” measure volume only. 10 units on a U-100 syringe = 0.1mL. This has nothing to do with IU of hormones or other biologics.
Shaking the vial. Vigorous shaking creates foam and mechanical stress that denatures peptide bonds. Always swirl gently in a circular motion until the powder dissolves completely.
Spraying water directly onto the powder. Direct water pressure disrupts the lyophilised cake. Instead, tilt the vial and let BAC water run slowly down the inner wall to dissolve the powder from the sides.
Using sterile water for multi-dose vials. Sterile water contains no preservative. Using it for repeated-access vials creates bacterial contamination risk. Use bacteriostatic water for any vial you will access more than once.
Adding the wrong BAC water volume. Changing the BAC water volume changes the concentration and therefore the units per dose. Always re-calculate units if you use a different water volume than planned.
Failing to label the vial. An unlabelled vial is a safety hazard. Label with: peptide name, date mixed, concentration (mcg/mL), and your initials. Discard any unlabelled reconstituted vials.
Storing in the refrigerator door. Door temperatures fluctuate every time the fridge opens. Store peptide vials in the main body of the refrigerator where temperature remains stable at 2–8°C.
Using a vial beyond 30 days. Even if solution remains in the vial, most reconstituted peptides degrade meaningfully after 28–30 days. Discard on or before the 28-day mark from the reconstitution date.
Not swabbing the septum before drawing. Always swab the rubber septum with a fresh alcohol swab before each draw, even if the vial was recently opened. Allow to air-dry before piercing.
