What It Is

Oxytocin binds oxytocin receptors (OTR) distributed throughout the brain (amygdala, hippocampus, brainstem, nucleus accumbens), periphery (uterus, mammary gland, heart, kidney). In the CNS, OTR activation in the amygdala reduces fear and anxiety by dampening amygdala threat responses. In the nucleus accumbens it enhances social reward and bonding. It modulates serotonin and dopamine release, contributing to mood and social motivation. Peripherally, it causes uterine contraction, milk letdown, and has cardioprotective effects. Intranasal administration achieves significant CNS penetration via olfactory neurons, bypassing the blood-brain barrier.

Supplies Needed

Sterile saline for injection

Reconstitution solvent

Nasal spray bottle (metered dose)

Intranasal administration device

1 mL syringes for subcutaneous use

Subcutaneous injection

Storage

Lyophilized (powder)

Store at 2-8C. Very temperature sensitive. Do not freeze.

Reconstituted (liquid)

Refrigerate at 2-8C; use within 14 days. Very short stability.

Dosing Protocols

Dose10-20 IU intranasal
FrequencyAs needed
Cycle LengthAs needed (not daily chronic use)
RouteIntranasal

Timing: 20-30 minutes before social interaction or intimacy. 10 IU = approximately 2 puffs of standard nasal spray; effects begin within 20-30 minutes

↓ Apply these values to the reconstitution calculator
Dose20-40 IU intranasal
FrequencyAs needed, maximum 1-2x daily
Cycle LengthAs needed
RouteIntranasal

Timing: 20-30 minutes before social or intimate activity. Standard research dose for social bonding and anxiety reduction; higher doses do not produce proportionally greater effects (inverted-U dose-response)

↓ Apply these values to the reconstitution calculator
Dose24 IU intranasal (subcutaneous 1-2 IU for systemic effects)
FrequencyAs needed
Cycle LengthAs needed; avoid daily chronic use
RouteIntranasal or subcutaneous

Timing: 30 minutes before intended effect. Some research uses subcutaneous injection for more reliable CNS delivery vs intranasal variability

Weekly Timeline

Week Expected Effects
Week 1 Acute effects within 30 minutes of each dose: enhanced social warmth, reduced anxiety, improved sexual arousal and intimacy
Week 4 With regular use: cumulative improvements in social connectivity and relationship quality; improved baseline anxiety

Injection Technique & Reconstitution

  1. Oxytocin is typically available as a nasal spray (Syntocinon) or solution for injection
  2. For lyophilized powder: Remove flip-top cap from vial
  3. Swab rubber stopper with alcohol swab
  4. Draw sterile saline into syringe
  5. Inject slowly down the vial wall and swirl gently
  6. For intranasal: transfer to metered nasal spray bottle
  7. Label with date; refrigerate at 2-8C
